Mutation alters the consensus 5' splice site of the second intron from GT to GA. If no splicing occurs, then because of stop codons in the intron, the protein product would terminate after 2001 amino acids and would be missing the C terminal 143 amino acids.

ash114/Df(3L)kto2 lethality occurs during late pupal or pharate stages.

Significant viability when raised at 20oC, with reduced viability at higher temperatures. Temperature-sensitive period for viability is primarily during the third larval instar. Exposure to 29oC for as little as one day during the third larval instar causes a high frequency of homeotic transformations of the third thoracic segment to the second thoracic segment with no significant loss of viability. ash11/ash114 or ash114/ash128 females show a reversible block in oogenesis at 27oC: an increasing number of stage 14 egg chambers is evident per ovary with increasing temperature. Pole cell transplantation reveals that embryogenesis requires germ line autonomous, maternal expression of ash1.
