DNA POLYMERASE ALPHA-PRIMASE -
DNA polymerase alpha-primase synthesizes short RNA-DNA primers on the lagging strand during DNA synthesis. It is a heterotetramer composed of two polymerase subunits (DNApol-α180 and DNApol-α73) and two primase subunits (DNApol-α50 and DNApol-α60). DNApol-α180 and DNApol-α50 encode the DNA polymerase and RNA primase catalytic activities, respectively, while DNApol-α73 and DNApol-α60 encode the regulatory subunits. (Adapted from PMID:30005324.)
RNA POLYMERASE CATALYTIC SUBUNITS -
DNA-directed RNA polymerases (RNAPs) catalyze the synthesis of RNA from DNA templates. Nuclear DNA is transcribed by three different multi-subunit polymerases, RNAPs I, II and III, which synthesize mRNAs, rRNAs, tRNAs and other non-coding RNAs. Although composed of between 12-17 subunits, all three nuclear RNAPs contain a ten-subunit catalytic core. Mitochondrial RNA is synthesized by a single subunit RNA polymerase. DNA primases are a class of RNA polymerases that catalyse the synthesis of short RNA primers that are extended by DNA polymerases during DNA replication. Protein Function (UniProtKB)
Catalytic subunit of the DNA primase complex and component of the DNA polymerase alpha complex (also known as the alpha DNA polymerase-primase complex) which play an essential role in the initiation of DNA synthesis (PubMed:6403945, PubMed:6409898, PubMed:6773966, PubMed:6806812, PubMed:6812052). During the S phase of the cell cycle, the DNA polymerase alpha complex (composed of a catalytic subunit PolA1, an accessory subunit PolA2 and two primase subunits, the catalytic subunit Prim1 and the regulatory subunit Prim2) is recruited to DNA at the replicative forks (By similarity). The primase subunit of the polymerase alpha complex initiates DNA synthesis by oligomerising short RNA primers on both leading and lagging strands (By similarity). These primers are initially extended by the polymerase alpha catalytic subunit and subsequently transferred to polymerase delta and polymerase epsilon for processive synthesis on the lagging and leading strand, respectively (By similarity). In the primase complex, both subunits are necessary for the initial di-nucleotide formation, but the extension of the primer depends only on the catalytic subunit (By similarity). Can add both ribo- and deoxynucleotides during elongation of the primers (PubMed:6812052). Binds single stranded DNA (By similarity).

          Subunits of DNA polymerase α and δ are purified from embryos and characterised.

          DNApol-α50 has been cloned and characterised. Expression patterns of DNApol-α50 protein and transcript throughout development are consistent with a role of the protein in DNA replication.

          The properties of the DNA polymerase α enzyme have been studied.

          25bp DNA oligomer containing m4T is used in a gel extension assay to measure the efficiency of incorporation of dATP and dGTP opposite m4T using a DNA polymerase α-primase complex. The DNA polymerase α shows a clear preference for pairing with dGTP.
