Vps609.2, Ist151 double mutant wing disc clones raised at 29[o]C bulge out of the disc epithelium, fail to exhibit multivesicular bodies and instead exhibit enlarged endosomes and large vesicles that contain membraneous structures and electro-dense, but do not show cell polarity defects (assessed by DE-Cad and Dlg localization in immuno-stainings, and by Zonula adherens and septate junction integrity in TEM). When reared at 18[o]C, however, these double mutants are cell lethal in the wing disc, as no clones are obtained.
