Description

Deletion within Vps60 that removes the coding sequence downstream of exon 2. The predicted truncated protein lacks 154 of the 226 amino acid residues of wild-type Vps60. Generated by imprecise excision of P{EP}Vps60G4547. The deletion does not affect the adjacent downstream Eip74EF gene (determined by complementation analysis).

The rare Vps609.2 homozygous third instar larvae that survive when raised at 18[o]C develop melanotic tumors and their wing imaginal discs are very small, as compared to controls. In larvae reared at 25[o]C until the end of L2 and then shifted to 18[o]C until the end of L3, wing discs are larger than those kept constantly at 25[o]C and also larger than discs of wild type controls. Multivesicle bodies in wing disc cells are significantly larger than in wild-type controls, regardless of temperature, and contain more electron-dense material.

Vps609.2 wing disc clones do not show cell polarity defects (assessed by DE-Cad and Dlg localization in immuno-stainings, and by Zonula adherens and septate junction integrity in TEM).

Vps609.2, Ist151 double mutant wing disc clones raised at 29[o]C bulge out of the disc epithelium, fail to exhibit multivesicular bodies and instead exhibit enlarged endosomes and large vesicles that contain membraneous structures and electro-dense, but do not show cell polarity defects (assessed by DE-Cad and Dlg localization in immuno-stainings, and by Zonula adherens and septate junction integrity in TEM).When reared at 18[o]C, however, these double mutants are cell lethal in the wing disc, as no clones are obtained.
