CRISPR construct designed to knockout the CDC50 gene when combined with a source of Cas9 endonuclease. Constitutively expresses two sgRNAs, both of which target coding exon sequence that is shared by all CDC50 splice isoforms.

The knockout of CDC50 in class IV dendritic arborizing neurons by the combination of CDC50sgRNA.U6 and Spyo\Cas9ppk.PS leads to some dendrite debris being detected on the underlying epidermal cells, but does not affect dendrite density or dendrite dendrite field size.

The combined tissue-specific knockout of CDC50 (CDC50sgRNA.U6+Spyo\Cas9ppk.PS) and expression of Mmus\Ano6D430G-L.lexAop.Tag:FLAG under the control of Ecol\lexAppk.PU leads to higher levels of dendrite debris from class IV dendritic arborizing neurons being detected on the underlying epidermal cells than upon either CDC50 knockout or Mmus\Ano6D430G-L.lexAop.Tag:FLAG expression; this phenotype is more severe upon heat shock. This "double mutant" background leads to significant vesicle shedding by class IV dendritic arborizing neurons compared to controls, which is suppressed by drprindel homozygosity.

The combined tissue-specific knockout of CDC50 (CDC50sgRNA.U6+Spyo\Cas9ppk.PS) and expression of Avic\GFPsfGFP.UAS.Tag:mLactC1C2 under the control of Scer\GAL4DcG leads to short dendrites in class IV dendritic arborizing neurons, as compared to CDC50 knockout alone; this phenotype is suppressed by drprindel homozygosity.

The tissue-specific double knockout of CDC50 and ATP8A (CDC50sgRNA.U6+ATP8AsgRNA.U6+Spyo\Cas9ppk.PS) in class IV dendritic arborizing neurons does not affect dendrite density or dendrite dendrite field size, as compared to controls.
