The expression of PsndsRNA.shRNA.Scer\UAS.2 under the control of Scer\GAL4Act5C.PU leads to early- to mid-pupal lethality, as compared to control. Expression under the control of Scer\GAL4bbg-C96 leads to wing defects, ranging from severe wing vein and blistering phenotypes to wing margin loss, as compared to controls. Constitutive expression of PsndsRNA.shRNA.Scer\UAS.2 under the control of Scer\GAL4elav-C155 leads to pupal lethality in males, semi-lethality in females, and the surviving adult females show a severely decreased lifespan, severe locomotor defects in negative geotaxis assays, wrinkled wings in a third of the population and a rough eye phenotype, as compared to controls; adulthood-only expression (temporally-controlled by Gal80[ts]) leads to no obvious defects in gender ratio or in the eye, but leads to a significant decrease in adult lifespan, age-dependent decrease in adult locomotion (significant decreases in negative geotaxis assay score at day 50, but not at days 10 and 30 post eclosion), age-dependent increase in the number of vacuoli in the adult brain (significant and progressive increase from day 30 until day 50 post eclosion) and to a significant increase in the number of apoptotic cells in 30 days post eclosion brains, as compared to controls.

The adulthood-only co-expression of PsndsRNA.shRNA.Scer\UAS.2 and NctdsRNA.shRNA.Scer\UAS.2 under the combined control of Scer\GAL4elav-C155 and Gal80[ts] enhances the decrease in adult lifespan and the increase in the number of vacuoli, but not of apoptotic cells, in the adult brain, as compared to the adulthood-only expression of PsndsRNA.shRNA.Scer\UAS.2 alone or NctdsRNA.shRNA.Scer\UAS.2 alone.

Xenogenetic Interactions
Statement
Reference

Both the pupal lethality in males and the rough eye and wrinkled wing phenotypes in females observed upon the expression of PsndsRNA.shRNA.Scer\UAS.2 under the control of Scer\GAL4elav-C155 are not suppressed by the co-expression of Hsap\PSEN1Scer\UAS.VTR.

Comments

The pupal lethality in males and the semi-lethality in females observed upon the expression of PsndsRNA.shRNA.Scer\UAS.2 under the control of Scer\GAL4elav-C155 are partially suppressed by the co-expression of Psn+14.Scer\UAS; while the rough eye and wrinkled wing phenotypes in adults females are also partially rescued by the Psn+14.Scer\UAS expression, the rescued adult males now present these phenotypes, as compared to controls.
