Detailed Description

rawjj102 mutant clones in class IV dendritic arborizing neuron (Cd4a) neurons exhibit defects in dendrite stability, with frequency of retraction matching growth. Terminal dendrites are short, mostly unbranched and exhibit reduced complexity in distal regions of the arbor. The average terminal dendrite length is significantly reduced. Terminal dendrite orientation is altered and they often grow in proximity to primary dendrites, rather than growing away from them. Dendrite-dendrite crossing events are common.

Homozygous rawjj102 mutants arrest as embryos with dorsal closure defects.

rawjj102 mutant terminal dendrites are enriched in F-actin compared to controls. The microtubule cytoskeleton is unaffected.

Larvae doubly heterozygous for rawjj102 and trc1 exhibit significant increased in dendrite-dendrite crossing in class IV dendritic arborizing neurons. Neither heterozygote alone has a significant effect on the number of crossing events. Dendrite number is unaffected.

Expression of trcScer\UAS.cUa under the control of Scer\GAL4109(2)80 enhances the number of dendrite-dendrite crossings seen in rawjj102 mutant class IV dendritic arborizing neuron clones.

Expression of trcScer\UAS.T:Myr-Src64B under the control of Scer\GAL4109(2)80 suppresses the number of dendrite-dendrite crossings seen in rawjj102 mutant class IV dendritic arborizing neuron clones.

Expression of trcT453E.Scer\UAS under the control of Scer\GAL4109(2)80 partially suppresses the number of dendrite-dendrite crossings seen in rawjj102 mutant class IV dendritic arborizing neuron clones.

Co-expression of mysScer\UAS.cDa and mewScer\UAS.cMBa under the control of Scer\GAL4elav-C155 suppresses the number of dendrite-dendrite crossings seen in rawjj102 mutant class IV dendritic arborizing neuron clones. Terminal branch number is significantly restored and a greater fraction of stable terminals are seen compared. However, the mutant terminal dendrite elongation defect is not rescued.

Larvae doubly heterozygous for rawjj102/AGO1jj472 class IV arborizing dendritic neuron clones have shorter terminal dendrites and less complex terminal arbors than controls. The frequency of dendrite-dendrite crossing over is similar to controls.

Comments

Expression of rawScer\UAS.short.T:Avic\GFP-EGFP under the control of Scer\GAL4109(2)80 fully rescues the dendrite defects seen in rawjj102 mutant class IV dendritic arborizing neuron clones.
