Description

The 520 amino acids corresponding to the third proline-rich region and the first CC motif of sick have been deleted. The non-coding genes CR43828, CR43827 and CR45183 are also deleted.

Comment:

Scer\FRT-mediated recombination between the two progenitor insertions has resulted in the deletion of the genomic sequence between them.

Homozygous sickΔ mutant mushroom bodies display short α/β axonal lobes compared with those of controls. No axonal lobe phenotypes are observed in the mushroom bodies of sickΔ/+ mutants.

The axons of single sickΔ mutant α/β mushroom body neurons fail to extend their branched axons to the lobe terminus.

sickΔ mutant clones in the core region of the α/β lobe of the mushroom body show slightly increased F-actin levels compared with wild type controls.

sickA384/sickΔ mutant mushroom bodies display short α/β axonal lobes compared with those of controls, although the morphology of the peduncle appears normal. In addition, sickA384/sickΔ mutants display defective ellipsoid body ring structures in each hemisphere of the central brain. A prominent increase in F-actin signal is detected in the core region of the defective α/β axonal lobe compared to controls.

sickΔ/Df(2L)ED1303 mutant mushroom bodies display short α/β axonal lobes compared with those of controls.

sickΔ/sickΔ-L1 mutant mushroom bodies display short α/β axonal lobes compared with those of controls.

sickΔ/sickΔ-L2 mutant mushroom bodies display short α/β axonal lobes compared with those of controls.

sickΔ/sickΔ-L3 mutant mushroom bodies display short α/β axonal lobes compared with those of controls.

sickΔ/sickΔ-2 mutant mushroom bodies display short α/β axonal lobes compared with those of controls.

sickΔ tsrN96A double heterozygotes do not show obvious axonal growth defects in α/β axonal lobes, peduncle and ellipsoid body.

The mushroom body axons of flies that are heterozygous for Rac1J11, Rac2Δ tsrN96A sickΔ and MtlΔ fail to extend to form peduncles and lobe structures (the 'posterior arrest' phenotype).

Expression of tsrScer\UAS.cNa under the control of Scer\GAL4NP7175 significantly suppresses the axon growth phenotype seen in the α/β mushroom body lobes of sickA384/sickΔ mutants.

Expression of tsrS3A.Scer\UAS under the control of Scer\GAL4NP7175 significantly suppresses the axon growth phenotype seen in the α/β mushroom body lobes of sickA384/sickΔ mutants.

Expression of tsrS3E.Scer\UAS under the control of Scer\GAL4NP7175 does not significantly suppresses the axon growth phenotype seen in the α/β mushroom body lobes of sickA384/sickΔ mutants.

Expression of sshScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4NP7175 partially suppresses the axon growth phenotype seen in the α/β mushroom body lobes of sickA384/sickΔ mutants.

Expression of sshCS.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4NP7175 does not significantly suppress the axon growth phenotype seen in the α/β mushroom body lobes of sickA384/sickΔ mutants.

Expression of sshScer\UAS.T:Ivir\HA1 suppresses the axon growth defect and F-actin elevation seen in the α/β mushroom body lobes when LIMK1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4ey-OK107. This suppression does not occur in a sickΔ/sickA384 mutant background.

Expression of Rac1Y40C.Scer\UAS.T:Hsap\MYC suppresses the axon growth defect and F-actin elevation seen in the α/β mushroom body lobes when LIMK1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4ey-OK107. This suppression does not occur in a sickΔ/sickA384 mutant background.

Expression of sickScer\UAS.cAa under the control of Scer\GAL4elav.PU almost fully rescues the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants. The rescued flies exhibit a β lobe fusion phenotype.

Expression of sickScer\UAS.cAa under the control of Scer\GAL4ey-OK107 significantly rescues the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.

Expression of sickScer\UAS.cAa under the control of Scer\GAL4NP7175 significantly rescues the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.

Expression of sickScer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4elav.PU almost fully rescues the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.

Expression of sickScer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4ey-OK107 significantly rescues the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.

Expression of sickScer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4NP7175 significantly rescues the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.

Expression of sickK1881A.Scer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4ey-OK107 fails to rescue the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.

Expression of sickK1881A.Scer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4NP7175 fails to rescue the axonal growth defects seen in the α/β mushroom body lobes of sickΔ/sickA384 mutants.
