Scer\FRT-mediated recombination between two progenitor insertions, resulting in a deletion that removes upstream coding exons of the current wake annotation (Annotations CG6954 and CG42686 were merged as CG45058 (wake) in release 5.53 of the genome annotation).
Homozygous flies show a significant reduction in total daily sleep and a significant increase in sleep latency compared to controls.
The action potential firing rate of l-LNv neurons in wakeD2 mutants is similar to that of controls at ZT0-ZT2, but is nearly 4-fold higher than controls at ZT12-ZT14 (indicating that the cells are more excitable at the day/night transition). Resting membrane potential of the mutant l-LNv cells is not significantly different between ZT0-ZT2 and ZT12-ZT14.
wakeD2 has decreased sleep | adult stage phenotype, non-enhanceable by Gat33-1
wakeD2 has abnormal sleep | adult stage phenotype, non-enhanceable by Gat33-1
wakeD2 has abnormal sleep phenotype, suppressible by Pdfrunspecified
wakeD2 has abnormal sleep phenotype, suppressible by Pdf01
wakeD2 has decreased sleep | adult stage phenotype, non-suppressible by Gat33-1
wakeD2 has abnormal sleep | adult stage phenotype, non-suppressible by Gat33-1
Rdl1, wakeD2 has abnormal sleep | dominant phenotype
The increase in sleep latency seen in wakeD2 flies is blocked if they are also mutant for either Pdfrunspecified or Pdf01.
Rdl1/wakeD2 double heterozygotes show a significant increase in sleep latency compared to controls (this phenotype is not seen in either single heterozygote).
