Description

Imprecise excision of the progenitor insertion, resulting in a 3062bp deletion (coordinates 2L:19766024..19769085 , release 5 genome) which extends from 156bp upstream of the first bsh start codon to 336bp downstream of the bsh stop codon.

In bsh1/In bsh2 mutants, there is strong disorganization of the columnar structures in the larval and pupal medulla.

MARCM neuron clones in the medulla that would normally express bsh (labelled using Scer\GAL4bsh.PH) but which are homozygous for bsh1 are transformed into transmedullary neuron (Tm)-type neurons in 89.6% of cases (bsh-expressing neurons normally develop into Mi1 medulla intrinsic neurons in wild-type animals).

MARCM neuron clones in the lamina that would normally express bsh (labelled using Scer\GAL4bsh.PH) but which are homozygous for bsh1 develop into L3-like neurons in 91.7% of cases (bsh is normally expressed in L4 and L5 lamina neurons in wild-type).

Expression of bshScer\UAS.PB under the control of Scer\GAL4bsh.PH partially rescues the transformation of Mi1 medulla intrinsic neurons into transmedullary neuron (Tm)-type neurons seen in bsh1 MARCM neuron clones. The projections of the rescued neurons are restricted to the medulla and arborisations are formed at the M1, M5 and M9-M10 layers, but the size and shape of the arborisations is not identical to wild-type Mi1 neurons.

Expression of bshScer\UAS.PA under the control of Scer\GAL4bsh.PH mildly rescues the transformation of Mi1 medulla intrinsic neurons into transmedullary neuron (Tm)-type neurons seen in bsh1 MARCM neuron clones.

Expression of bshScer\UAS.PB under the control of Scer\GAL4bsh.PH partially rescues the formation of ectopic L3-like lamina neurons seen in bsh1 MARCM neuron clones.
