Homozygotes and RhoGAP92B1/RhoGAP92B2 animals have no obvious defects in pathfinding of motor axons.

Third instar RhoGAP92B1/RhoGAP92B2 larvae show a mild increase in muscle area. They show a 43% reduction in bouton number/muscle area and a 37% reduction in neuromuscular junction (NMJ) length/muscle area compared to wild type. Mean bouton size is increased by 69% compared to controls. Muscle area and NMJ length are normal in first instar RhoGAP92B1/RhoGAP92B2 larvae.

Ultrastructurally, type I boutons show a number of defects in RhoGAP92B1/RhoGAP92B2 third instar larvae. The cross-sectional bouton area is increased and the number of active zones/bouton area is reduced, while the number of T bars/bouton area is normal. Subsynaptic reticulum (SSR) thickness/bouton area is normal, SSR density is reduced and total non-tubular sarcoplasmic area/SSR area is increased.

The amplitude of the excitatory junctional current (EJC) is reduced in RhoGAP92B1/RhoGAP92B2 third instar larvae compared to wild type. The amplitude of the miniature EJC is normal, but there is a small but significant increase in mEJC frequency.

Expression of gbbScer\UAS.T:Avic\GFP under the control of Scer\GAL4C57 partially suppresses the reduction in bouton number/muscle area and satellite bouton number at the neuromuscular junction seen in third instar RhoGAP92B1/RhoGAP92B2 larvae.

The electrophysiological defects seen in third instar RhoGAP92B1/RhoGAP92B2 larvae are not rescued by expression of RhoGAP92BScer\UAS.cNa under the control of Scer\GAL4elav-C155 but are completely rescued by expression of RhoGAP92BScer\UAS.cNa in the muscle under the control of Scer\GAL4C57.

The increased bouton size, increased bouton area and reduced number of active zones/bouton area are rescued, but the subsynaptic reticulum defects are not rescued.
