Flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency) are arrhythmic for locomotor activity under constant darkness conditions.

Circadian behaviour under 12 hour light/12 hour dark conditions is altered in flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency) The morning peak of activity is severely blunted, although a robust evening peak is still present. The phase of the evening peak of activity is advanced compared to control flies.

Flies expressing gwHMS00105 under the control of Scer\GAL4P2.4.Pdf show normal circadian locomotor behaviour under constant darkness conditions.

Co-expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 rescues rhythmic locomotor behaviour under constant darkness conditions in 41.0% of flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency).

Locomotor activity behaviour under 12 hour light/12 hour dark conditions is partially rescued in flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa) if they are also mutant for dnc1. The evening peak phase is rescued closer to a wild type value, while the morning peak is not restored. 41.0% of flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa) in a dnc1 background are rhythmic for locomotor activity under constant darkness conditions.

Comments

Co-expression of gwshRNAres.Scer\UAS restores rhythmic locomotor behaviour under constant darkness conditions flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency). The rescued flies often have a long period phenotype. Under 12 hour light/2 hour dark conditions, both the morning peak and the evening peak are normal in phase and amplitude in the rescued flies.

Co-expression of gwAA.shRNAres.Scer\UAS usually fails to restore rhythmic locomotor behaviour under constant darkness conditions in flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency). The locomotor activity defects seen under 12 hour light/2 hour dark conditions are not rescued.
