UASt regulatory sequences drive expression of gw containing a number of synonymous codon mutations such that the binding site for the shRNA encoded by P{TRiP.HMS00105} has been extensively mutated without effecting the amino acid sequence. In addition, the N-terminal glycine-tryptophan (GW) motifs critical for AGO1 binding have been mutated to alanine residues (AA).
Scer\GAL4tim.PE/gwAA.UAS.RR fails to rescue gwHMS00105
Co-expression of gwAA.shRNAres.Scer\UAS usually fails to restore rhythmic locomotor behaviour under constant darkness conditions in flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency). The locomotor activity defects seen under 12 hour light/2 hour dark conditions are not rescued.
