UAS regulatory sequences drive expression of wild-type gw containing a number of synonymous codon mutations such that the binding site for the shRNA encoded by P{TRiP.HMS00105} has been extensively mutated without effecting the amino acid sequence.
Co-expression of gwshRNAres.Scer\UAS restores rhythmic locomotor behaviour under constant darkness conditions flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency). The rescued flies often have a long period phenotype. Under 12 hour light/2 hour dark conditions, both the morning peak and the evening peak are normal in phase and amplitude in the rescued flies.
