Expression of Sas-6Ubi.P.GFP does not lead to the formation of the procentriolar ectopic particles in mature primary spermatocytes when compared to controls.

Overexpression of Sas-6Ubi.P.T:Avic\GFP leads to some centriole overduplication in third instar larval brain cells.

The overexpression of Sas-6Ubi.P.T:Avic\GFP does not cause centriole overduplication in spermatocytes.

Overexpression of Sas-6Ubi.P.T:Avic\GFP in syncytial embryos results causes approximately 18% of centrioles to overduplicate.

Unfertilized eggs laid by mothers carrying two copies of sas-6Ubi.P.T:Avic\GFP assemble centriole-like structures.

Approximately 3% of centrioles proceed through an extra round of template-driven replication within a single nuclear cycle in syncytial embryos expressing sas-6Ubi.P.T:Avic\GFP, and a few centrioles undergo two extra rounds of replication.

Expression of ana2Ubi.C.GFP in combination with Sas-6Ubi.P.GFP leads to the formation of the procentriolar ectopic particles that are located in the cytoplasm of mature and immature primary spermatocytes, and attached to the centrioles only in mature primary spermatocytes when compared to controls. Expression of Poc1UAS.B.GFP (under the control of Scer\GAL4VP16.bam) together with Sas-6Ubi.P.GFP but not with Poc1UAS.A.GFP or Poc1BΔC.UAS.GFP leads to the formation of the procentriolar ectopic particles in mature primary spermatocytes when compared to controls. Procentriolar ectopic particles in mature primary spermatocytes resemble the proximal centriole-like ultrastructure with tubule-like structures (significantly wider in Poc1 Sas-6 than Ana2 Sas-6 co-expression), spokes (only in Ana2 Sas-6 co-expression), and electron-dense material (similar diameter in both Poc1 Sas-6 than Ana2 Sas-6 co-expression).

Expression of ana2Ubi.C.GFP in combination with Sas-6Ubi.P.GFP leads to formation of procentriolar ectopic particles while their expression in Poc1W87X homozygous background leads to structural defects in procentriolar ectopic particles such as fragmented tubule-like structures and electron-dense material in mature primary spermatocytes.
