Expression of smGD12545 under the control of Scer\GAL4GMR.PU has no obvious effect on adult eye morphology, whereas expression driven by Scer\GAL4elav.PU results in significant decrease in adult climbing ability.

Adults expressing smGD12545 under the control of the cardioblast-specific Scer\GAL4tin.CΔ4 driver show significantly reduced survival on day 6 after a shift to 29[o]C compared to control flies.

Adults expressing smGD12545 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4Mef2.PR results in grossly normal adult indirect flight muscles.

Expression under the control of Scer\GAL4Mef2.PR results in degenerate myofibrils in the adult indirect flight muscles.

Expression under the control of Scer\GAL4Mef2.PR results in sarcomeres with broadened Z-lines in the adult indirect flight muscles.

Expression under the control of Scer\GAL4pnr-MD237 may result in semi-lethality, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in loss of bristles on the notum in 0% or 40-50% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in bristle morphology defects on the notum in 0% or 20-30% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Statement

The climbing ability deficit observed both in the TBPH hypomorphic flies (TBPHΔ23/+;Scer\GAL4elav.PU>TBPHdsRNA.Scer\UAS.cUa, UAS-Dicer2) and flies expressing smGD12545 (also with UAS-Dicer2 and under Scer\GAL4elav.PU control) is much enhanced when the two mutants are combined.

The eye degeneration phenotype (appearance of melanotic spots, fusion of ommatidia) characteristic for adult flies expressing TBPHScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU is mildly suppressed by co-expression of smGD12545.
