Flies expressing qsmGD4817 under the control of Scer\GAL4tim.PE exhibit robust, long-period locomotor rhythms in constant dim white light, in contrast to control flies which largely show arrhythmic behavior in these conditions. These flies do not exhibit any difference in locomotor rhythms when kept in constant darkness, as compared to controls.

The l-LNv neurons in flies expressing qsmGD4817 under the control of Scer\GAL4Pdf.PU exhibit depolarization of the nighttime (but not daytime) resting membrane potential, and an increased spontaneous firing rate during the night (but not during the day), as compared to wild type; these neurons also show a heightened response to blue light when tested during the daytime, as compared to wild type.

The l-LNv neurons in flies expressing qsmGD4817 under the control of Scer\GAL4tim.PE exhibit an increased spontaneous firing rate as compared to wild type.

Under dim (35-55 lux) constant light conditions, 93.3 to 100% of flies expressing qsmdsRNA.Scer\UAS under the control of qsmGD4817 show robust locomotor activity rhythmicity, with an average period ranging from 26.2 or 27.9 hours (depending on the lines used), whereas control animals are largely arrhythmic under these conditions.

Adults expressing qsmGD4817 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Co-expression of ShawRA.Scer\UAS.T:Zzzz\FLAG suppresses, or of NKCCNIG.2509R partially suppresses the increased spontaneous firing rate of l-LNv neurons in flies expressing qsmGD4817 under the control of Scer\GAL4tim.PE.
