Expression of LamGD1478 in the fat body under the control of Scer\GAL4Cg.PA results in increased midgut hyperplasia compared to controls. This increase in hyperplasia is weaker when the flies are maintained in axenic conditions, but is still increased compared to wild type flies in axenic conditions.

The nuclei of many cyst stem cells in the mutant gonad are positioned adjacent to hub cells in third instar larvae expressing LamGD1478 under the control of Scer\GAL4tj-NP1624 (in wild type the nuclei are dispersed away from the hub), while the number of germline stem cells associated with the hub is decreased compared to wild type.

The cell bodies of cyst cells expressing LamGD1478 under the control of Scer\GAL4tj-NP1624 remain compacted in the developing testis (in wild type they expand to surround the germline cyst).

Adults expressing LamGD1478 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4Mef2.PR results in late pupal lethality.

Expression under the control of Scer\GAL4pnr-MD237 results in loss of bristles on the notum in 50-60% or 70-80% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in bristle morphology defects on the notum in 0% or 20-30% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in planar polarity defects in the orientation of bristles of the notum in 0% or 30-40% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 0% or 30-40% of the Scer\GAL4pnr-MD237-expressing area of the notum, depending on the insertion line used.

Expression of one copy of Su(var)205hs.T:Ecol\lacI-DNA-BD partially suppresses the midgut hyperplasia seen when LamGD1478 is expressed under the control of Scer\GAL4Cg.PA.

Co-expression of rlScer\UAS.T:Uuuu\nls6 partially rescues the germline stem cell differentiation defect seen in males expressing LamGD1478 under the control of Scer\GAL4tj-NP1624 : spermatocytes and sperm bundles are formed in 45.2% of testes and the increased number of early germ cells is partially reduced in the remaining testes. Fertility is not rescued.

Co-expression of LamCScer\UAS.cSa partially rescues the germline stem cell differentiation defect seen in males expressing LamGD1478 under the control of Scer\GAL4tj-NP1624 : 58.9% of testes show strong rescue (although they are shorter than normal), containing germ cells at all stages of differentiation. The remaining 41.1% of testes show weak rescue, showing less severe accumulation of early germ cells but still appearing abnormal.
