Clonal expression of ATP6AP2GD2871 under the control of Scer\GAL4αTub84B.PC in the third instar larval fat body, Malpighian tubule or pupal wing leads to an increase in lipid droplet size, as compared to the neighboring control tissue; ATP6AP2GD2871-expressing fat body clones also display an increase in the number of autophagosomes (identified as Atg8a-mCherry-positive puncta), as compared to neighboring control tissue.

Expression of VhaM8.9GD2871 driven by Scer\GAL4GMR.PF (together with UAS-dicer2 to enhance RNAi efficiency) does not lead to external eye morphology phenotypes, but flies show significant defects in phototaxis behavior compared to controls. Electroretinograms (ERGs) from Scer\GAL4GMR.PF>VhaM8.9GD2871 eyes reveal synaptic defects, with complete absence of 'on' or 'off' transients and diminished amplitude photoreceptor re- and depolarization in response to light.

Expression of VhaM8.9GD2871 driven by Scer\GAL4elav.PLu (together with UAS-dicer2 to enhance RNAi efficiency) does not result in lethality and flies show locomotor defects (less severe than those seen with expression of VhaM8.9KK107676). The area of synaptic terminals is significantly decreased and there is a significant increase in the number of satellite boutons (no significant difference in active zone number) at the neuromuscular junction in Scer\GAL4elav.PLu>VhaM8.9GD2871 third instar larvae, compared to controls; synapses also contain large multivesicular body-like organelles and T-bars at active zones are perturbed. Scer\GAL4elav.PLu>VhaM8.9GD2871 adult brains show impairments in autophagy.

Expression of VhaM8.9GD2871 under the control of Scer\GAL4nub-AC-62 at 25[o]C results in severe growth defects in the wing.

Expression of VhaM8.9GD2871 under the control of Scer\GAL4nub-AC-62 at 18[o]C results in loss of wing margin bristles and misorientation of wing hairs.

Expression of VhaM8.9GD2871 under the control of Scer\GAL4ap-md544 results in abnormal polarity of notum bristles.

Expression of VhaM8.9GD2871 under the control of Scer\GAL4ap-md544 results in severe planar polarity defects in the anterior-posterior orientation of sensory bristles or microchaetae of the notum. Abnormalities in bristle morphology and number are also seen.

Expression of VhaM8.9GD2871 under the control of Scer\GAL4dpp.blk1 results in cell autonomous planar cell polarity defects in the wing, including wing hair misorientation and multiple wing hairs in the proximal part of the wing. The wing compartment expressing VhaM8.9GD2871 under the control of Scer\GAL4dpp.blk1 is reduced in size and there is both loss of wing vein and ectopic wing vein material.

Expression of VhaM8.9GD2871 under the control of Scer\GAL4bbg-C96 results in an intermittent loss of sensory bristles at the wing margin.

Adults expressing VhaM8.9GD2871 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4Mef2.PR results in adult/pharate lethality.

Expression under the control of Scer\GAL4pnr-MD237 results in bristle morphology defects on the notum in 80-90% of the Scer\GAL4pnr-MD237 expression domain.

Expression under the control of Scer\GAL4pnr-MD237 results in planar polarity defects in the orientation of bristles of the notum in 80-90% of the Scer\GAL4pnr-MD237 expression domain.

The wing growth defects seen in animals expressing VhaM8.9GD2871 under the control of Scer\GAL4nub-AC-62 at 25[o]C are suppressed in 70% of flies by co-expression of Hsap\ATP6AP2Scer\UAS.T:Avic\GFP-EGFP.

The abnormal polarity of notum bristles caused by expression of VhaM8.9GD2871 under the control of Scer\GAL4ap-md544 is rescued by co-expression of Hsap\ATP6AP2Scer\UAS.T:Avic\GFP-EGFP.
