Detailed Description

santa-maria1 animals exhibit defects in the function of abdominal lateral pentascolopidial chordotonal organ lch5 (in third instar larvae) and Johnston organ (in adults) mechanoreceptors compared to controls.

santa-maria1 homozygous mutant third instar larvae show impaired temperature selection and unlike controls display no strong preference for cooler temperatures (18[o]C) in a temperature gradient. This effect is not light-sensitive and does not appear to be due to developmental delay as the mutants enter third instar stage with similar timing as controls.

Mechanical and electrical responses to sound in the Johnston's organ are impaired in mutant flies.

Mutant larvae show defects in discrimination between 18[o]C and 24[o]C in a binary choice thermotaxis assay compared to controls. This defect is reversed if all trans-retinal is added to the food.

Electroretinograms from santa-maria1 flies fail to show prolonged depolarization afterpotential (PDA) in response to blue light. This phenotype is rescued when flies are fed with all-trans-retinal but not when they are fed with β-carotene.

A santa-maria1 homozygous mutant background partially suppresses the Ras85DV12.Scer\UAS-induced overgrowth, including the aberrant histology of the salivary glands and the mortality.

Expression of santa-mariaScer\UAS.cWa under the control of Scer\GAL4ph-p-NP0761 rescues the defects in mechanical and electrical responses of the Johnston's organ to sound which are seen in santa-maria1 flies.

Expression of santa-mariahs.PW restores a prolonged depolarization afterpotential in response to blue light in electroretinogram recordings from santa-maria1 flies.
