Ubiquitous and constitutive overexpression of Ctr1BScer\UAS.cSa under the control of Scer\GAL4Act5C.PI leads to complete larval and pupal lethality when the flies are raised on normal food. The observed lethality is copper-dependent, since a large fraction of the flies successfully reach the adult stage if they are raised on food containing the copper-specific metal chelator BCS.

Overexpression of Ctr1BScer\UAS.cSa in the eye under the control of Scer\GAL4GMR.PF results in a rough eye phenotype in flies raised on normal food. This phenotype is strongly aggravated in flies raised on 100 υM copper. The strong rough eye phenotype is accompanied by high lethality. A 250υM BCS (a copper chelator) food supplement completely restores the wild-type eye phenotype and wild-type levels of survival.

Strong, ubiquitous expression of Ctr1BScer\UAS.cSa under the control of Scer\GAL4Act5C.PU is lethal when the flies are raised on normal food. Such flies can be partially rescued (<50%) by growing them on food supplemented with the copper chelator BCS.

Strong, eye-specific expression of Ctr1BScer\UAS.cSa under the control of Scer\GAL4GMR.PF leads to severely disrupted eye morphology (the rigid quasi-crystalline pattern of the ommatidia is lost and ommatidia are fused, leading to an uneven eye surface; in addition, overall eye shape is changed) when raised on normal food. This rough eye phenotype can be fully rescued by the addition of the copper chelator BCS.

Expression of Ctr1BScer\UAS.cSa under the control of Scer\GAL4GMR.PF results in a rough eye phenotype when the flies are raised on food supplemented with 20μM copper.

Flies with the Ctr1BScer\UAS.cSa transgene, under the regulation of Scer\GAL4Act5C are lethal. In the presence of bathocuproinedisulfonate (a copper chelator), these flies survive.

Overexpression of Ctr1BScer\UAS.cSa under the control of Scer\GAL4tub fails to suppress the lethality associated with developmental arrest found in Ctr1A25 mutants. This failure could be associated with toxicity caused by excessive copper uptake.

The rough eye phenotype caused by expression of Ctr1BScer\UAS.cSa under the control of Scer\GAL4GMR.PF in the presence of 20μM copper is suppressed by co-expression of one of MTF-1Scer\UAS.cEa, MtnAScer\UAS.cEa or MtnBScer\UAS.cEa, but is not suppressed by co-expression of one of MtnCScer\UAS.cEa or MtnDScer\UAS.cEa.

Constitutive expression of Ctr1BScer\UAS.cSa, under the regulation of Scer\GAL4Act5C rescues the developmental arrest and larval lethality of MTF-1140-1R mutants under copper depletion, with several viable MTF-1140-1R Ctr1BScer\UAS.cSa (Scer\GAL4Act5C) flies surviving on food containing 50 or even 100υM bathocuproinedisulfonate (a copper chelator). The rescued flies are normal and fertile.
