Description

UASt regulatory sequences drive expression of a membrane-targeted (by the CAAX motif) kinase-dead aPKC sequence. Carries the amino acid replacement K293W.

UASt regulatory sequences drive expression of a dominant-negative, kinase-dead form of aPKC (carries a K293W amino acid substitution in the ATP binding site) that is targeted to the membrane due to the presence of a Tag:CAAX(Unk) tag.

Expression of aPKCCAAXDN.Scer\UAS under the control of Scer\GAL4Bx-MS1096 has no effect on growth or survival of the wing discs.

Expression of aPKCCAAXDN.Scer\UAS under the control of Scer\GAL4wor.PA has no affect on the number of neuroblasts in the larval brain.

Overexpression of aPKCCAAXDN.Scer\UAS, under the control of Scer\GAL4en-e16E results in the loss of membrane associated crb and Patj concomitant with an apical expansion of basolateral markers.

Expression of aPKCCAAXDN.Scer\UAS under the control of Scer\GAL4Bx-MS1096 suppresses the tumorous growth seen in slmbUU11 mutant wing disc clones. The resulting clones are smaller than in wild type.

Coexpression of aPKCCAAXDN.Scer\UAS partially suppresses both the polarity and proliferation phenotypes of wing discs expressing Syx7dsRNA.Scer\UAS.cLa under the control of Scer\GAL4en-e16E.

Coexpression of aPKCCAAXDN.Scer\UAS partially suppresses the early pupal lethality associated with expression of Syx7dsRNA.Scer\UAS.cLa under the control of Scer\GAL4en-e16E, increasing survival of pharate adults from 30% to 91%.

Expression of kinase-dead aPKCCAAXDN.Scer\UAS, under the regulation of Scer\GAL4en-e16E largely normalises the effect caused by overexpression of wild-type crbintra.Scer\UAS (under the control of Scer\GAL4en-e16E). Expression of kinase-dead aPKCCAAXDN.Scer\UAS, under the regulation of Scer\GAL4en-e16E normalises the effect caused by overexpression of wild-type crbT6DT9D.intra.Scer\UAS (under the control of Scer\GAL4en-e16E) in approximately 40% of the flies, even though these flies still display strong cuticular defects.
