Expression of ineRA.Scer\UAS under the regulation of Scer\GAL4MZ1580 confers reduced excitability on the larval stage motor neuron, with a decreased rate of onset of long-term facilitation. Expression of ineRA.Scer\UAS under the regulation of Scer\GAL4MZ1580 confers temperature sensitive paralysis. 92% of flies carrying both ineRA.Scer\UAS and Scer\GAL4Gli-rL82 constructs become paralysed after transfer from 18oC to 38oC, whereas flies carrying either ineRA.Scer\UAS or Scer\GAL4Gli-rL82 do not show this paralysis. Only 4.5% of flies expressing ineRA.Scer\UAS under the regulation of Scer\GAL4MZ1580 become paralysed when placed at 29oC. Larvae in which ineRA.Scer\UAS is expressed under the control of either Scer\GAL4MZ1580 or Scer\GAL4Gli-rL82 exhibit a higher frequency of failures in evoked transmitter release from larval stage motor nerve terminals when bathed in varying Ca2+ concentrations. When ineRA.Scer\UAS is expressed under the control of Scer\GAL4Gli-rL82, at the permissive temperature of 21-22oC, larvae exhibit compound action potentials with a reduced amplitude compared to wild-type. At the restrictive temperature of 38oC, larvae expressing ineRA.Scer\UAS under the control of Scer\GAL4Gli-rL82 exhibit a complete loss of compound action potentials. This loss is reversed when the temperature is lowered to a permissive range.

*S Expression of ineRA.Scer\UAS driven by either Scer\GAL4Gli-rL82 or Scer\GAL4MZ1580 fully suppresses the downturned wings phenotype of Sh14 ine1. The Sh14 hyperexcitability phenotype, such as leg-shaking, can be suppressed by expression of ineRA.Scer\UAS under the control of Scer\GAL4MZ1580.

When ineRA.Scer\UAS is expressed under the control of Scer\GAL4Gli-rL82 in a para63 mutant, 73% of the flies become paralysed at 29oC.

At 32oC, all para63 flies expressing ineRA.Scer\UAS under the control of Scer\GAL4Gli-rL82 are paralysed.

These flies also require even more repetitive nerve stimulation compared to wild-type for the onset of long-term facilitation. Expression of ineRA.Scer\UAS driven by Scer\GAL4MZ1580 in the glia, results in 94% rescue of the Sh14 ine1 downturned wing phenotype. The Sh14 hyperexcitability phenotype, such as leg-shaking, can be suppressed by expression of ineRA.Scer\UAS under the control of Scer\GAL4MZ1580. When ineRA.Scer\UAS is expressed under the control of Scer\GAL4Gli-rL82, in a para63 mutant, 73% of the flies become paralysed at 29oC. When para63 flies, expressing ineRA.Scer\UAS under the control of Scer\GAL4Gli-rL82, are placed at 32oC, all the flies are paralysed.
