Description

The loxP cassette in the P{EP(loxP.UAS)y+.UAS} insertion has been excised, converting the inserted element to a single-headed P{EP(-loxP)y+.UAS} element capable of driving Rheb expression. in the presence of a GAL4 driver.

Fat body clones expressing RhebEP50.084-loxP under the control of Scer\GAL4Tub.PU, induced during embryogenesis and analyzed at the early third instar larval stage, bear larger cells than controls.

Clones expressing RhebEP50.084-loxP under the control of Scer\GAL4Act5C.PU in the third larval instar wing disc and fat body are larger than control clones.

Ovarian follicle cells overexpressing RhebEP50.084-loxP (under the control of Scer\GAL4Scer\FRT.Rnor\CD2.Act5C) lack lysotracker staining even under starvation conditions, indicating that activation of IIS/TOR signaling is sufficient to suppress starvation-induced autophagy.

Cells in the fat body that are expressing RhebEP50.084-loxP show an increase in relative cell area compared to controls.

Overexpression of RhebEP50.084-loxP under the control of Scer\GAL4796 induces a significant increase in the synaptic domain.

Expression of RhebEP50.084-loxP under the control of Scer\GAL4GMR.PF results in a bulging eye phenotype.

The reduction in the distance between wing veins 3 and 4 that is seen in animals expressing PrpkdsRNA.Scer\UAS.462 under the control of Scer\GAL4salm.EPv is not suppressed if they are also co-expressing RhebEP50.084-loxP.

Clones expressing PrpkdsRNA.Scer\UAS.462 and also co-expressing RhebEP50.084-loxP under the control of Scer\GAL4Act5C.PU in the third larval instar wing disc and fat body are small and apoptotic, showing the same phenotype as clones expressing PrpkdsRNA.Scer\UAS.462 alone under the control of Scer\GAL4Act5C.PU.

The increase in cell area relative to control cells that is seen in cells in the fat body that are expressing RhebEP50.084-loxP is enhanced if the animals are also homozygous for RagCEY11726.

The variable reduction in cell size and induction of autophagy in the fat body that is seen in animals expressing Atg1GS10797 under the control of Scer\GAL4Cg.PA is suppressed if the animals are also co-expressing RhebEP50.084-loxP.

The bulging eye phenotype caused by expression of RhebEP50.084-loxP under the control of Scer\GAL4GMR.PF is not suppressed by simultaneous coexpression of both scylEP22.1 and chrbEP1035.
