Expressing NiPp1UAS.EGFP under the control of either Scer\GAL4tim.UAS or Scer\GAL4Pdf.PU show lengthened circadian periods.

Files expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4P2.4.Pdf have a locomotor activity period approximately 1.5 hours longer than that of controls.

Files expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4tim.Scer\UAS have a locomotor activity period approximately 2.8 hours longer than that of controls. In addition, the amplitude of the circadian rhythm is reduced. These flies show long period rhythms that degenerate into arrhythmia after 4-6 days under constant darkness conditions.

The amplitude of phase shifts and the effective duration of the phase delay domain is increased in flies expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4tim.Scer\UAS compared to wild type when the phase response curve for light is examined. The phase delay-to-phase advance transition point occurs approximately 2 hours later than in controls, but the phase advance domains are about the same duration as in controls.

More than 95% of animals expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4arm.PS die either before or just after pupation. Squash preparations of third larval instar brains contain fewer cells per microscope field than normal and a significantly higher proportion of cells in mitosis than in wild type. Overcondensed chromosomes are seen. Indirect flight muscles are either not present or are in complete disarray in the few adult survivors expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4arm.PS. The jump muscle is mostly normal. Flies expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4vg.PM resemble wild type.

The indirect flight muscles are restored in adults expressing NiPp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4arm.PS if they are also expressing flwScer\UAS.T:Ivir\HA1. The modest wing overgrowth phenotype caused by expression of putScer\UAS.cMa under the control of Scer\GAL4vg.PM is enhanced by co-expression of NiPp1Scer\UAS.T:Ivir\HA1.
