Detailed Description

The average penetrance of ectopic eye induction in eyaScer\UAS.cHa; Scer\GAL4dpp.blk1 flies is 49% (over 8 independent P{UAS-eya.H} insertions). Ectopic eyes are typically seen on the head, either close to the eyes and near the base of the antennae. Patches of red pigement are also frequently seen in the wing hinges and on the underside of metathoracic legs, and ocassionally elsewhere on wings legs and antennae. The penetrance of ectopic eye induction in eyaScer\UAS.cHa; Scer\GAL4dpp.blk1 flies is reduced from 98% (n=298) to 50% (n=313) by heterozygosity for eyaG130 (data from a single, 'strong' P{UAS-eya.H} insertion). eyaScer\UAS.cHa; Scer\GAL4dpp.PH flies have highly variable wing defects, including ectopic bristles on L3 and in more extreme cases, blistering, and defects in overall shape: Proportion with blistered wings = 40%; proportion with only mild wing defects = 60%. These flies also have increased numbers of scutellar machrochaete (average= 6.0, compared to 4 in wild-type), and problems with thoracic closure. They also have a low penetrance of rough eyes, and arista to leg transformations. Note: all of the above data is an average result from 8 independent P{UAS-eya.H} insertions. Heterozygosity for eyaG130 leads to a decrease in the proportion of eyaScer\UAS.cHa; Scer\GAL4dpp.PH flies with blistered wings (76% to 3%) and an increase in the proportion with only milder wing defects (24% to 97%) (data from a single, 'strong' P{UAS-eya.H} insertion; n=approximately 300 in each case).

The penetrance of ectopic eye induction in eyaUAS.cHa; Scer\GAL4dpp.blk1 flies is reduced from 98% (n=298) to 50% (n=313) by heterozygosity for rlS-135 and to 58% (n=313), by heterozygosity for Egfrunspecified (data from a single, 'strong' P{UAS-eya.H} insertion). No suppression of penetrance was seen due to heterozygosity for bsk1 or the presence of rlSem.cUa.UAS, or bskUAS.cBa. Ectopic eyes in eyaUAS.cHa; Scer\GAL4dpp.blk1; rlSem.cUa.UAS are generally larger, and have more ommatidial definition than those in eyaUAS.cHa; Scer\GAL4dpp.blk1 flies. The proportion of eyaUAS.cHa; Scer\GAL4dpp.PH flies with blistered wings (76%) is decreased by heterozygosity for dac3 (to 4%), so3 (to 17%), rlS-135 (to 10%), or Egfrunspecified (to 14%), increased by heterozygosity for rlSem.cUa.UAS (to 93%), and relatively unaffected by heterozygosity for bsk1 (69%), or the presence of bskUAS.cBa (82%). Conversely, the proportion of eyaUAS.cHa; Scer\GAL4dpp.PH flies with only mild wing defects (24%) is increased by heterozygosity for dac3 (to 96%), so3 (to 83%), rlS-135 (to 90%), or Egfrunspecified (to 86%) decreased by heterozygosity for rlSem.cUa.UAS (to 7%), and relatively unaffected by heterozygosity for bsk1 (31%), or the presence of bskUAS.cBa (18%). (All data from experiments with a single, 'strong' P{UAS-eya.H} insertion; n=approximately 300 in each case).
