Detailed Description

CblKG03080 homozygous mutant third instar larvae have significantly increased concentration of circulating hemocytes compared to wild-type controls due to an increase in their mitotic activity (higher percentage of pH3-positive hemocytes). Primary hemocytes from these mutant larvae show strongly reduced internalization of sSpi-GFP protein (spi encodes the principal ligand of the EGFR pathway) at high sSpi-GFP conditions (treatment with 30 ng/ml sSpi-GFP), as compared to controls.

Homozygous CblKG03080 flies show poor survival to adulthood.

Less than half of CblEY11427/CblKG03080 mutant males survive to adulthood.

Pupation is delayed by approximately 24 hours in CblEY11427/CblKG03080 larvae compared with wild-type controls.

CblEY11427/CblKG03080 males show increased sensitivity to starvation, exhibiting an approximately 20% decrease in the median survival time upon food deprivation.

CblEY11427/CblKG03080 males display a dramatically shortened lifespan, with an approximately 67% reduction in the median lifespan compared with wild-type controls.

Heterozygotes show a direct effect on the shape of the wing.

Comments

The increased concentration of circulating hemocytes in CblKG03080 homozygous mutant third instar larvae along with the strongly reduced internalization of sSpi-GFP protein (spi encodes the principal ligand of the EGFR pathway) at high sSpi-GFP conditions (treatment with 30 ng/ml sSpi-GFP) seen in primary hemocytes isolated form the mutant larvae is fully rescued by expression of CblL.Scer\UAS but not CblL.Δ70Z.Scer\UAS under the control of Scer\GAL4crq.PO in the CblKG03080 mutants.
