Maternally and zygotically mutant arm043A01 embryos (coming from mothers whose whole germline constitutes from arm043A01 mutant cells only - created by the OvoD germline clone technique) fall apart during development due to loss of cell adhesion.

arm043A01 mutant embryos exhibit cuticle defects.

Epithelial tears occur in the ventral furrow of embryos mutant for maternal and zygotic arm043A01, but tears are not observed in more lateral ectoderm at the same stage. The mutant ventral cells appear to round up upon loss of adhesion. Despite the dramatic loss of adhesion, cells in maternal and zygotic arm043A01 mutant embryos maintain thin membrane connections, or tethers, to their neighbours.

Cuticle fails to form in embryos mutant for arm043A01 both maternally and zygotically.

arm043A01 clones deficient for both maternal and zygotic arm display gaps between furrows of cellularization embryos. Cell-cell adhesion is compromised and no detectable basal junctions form. However, furrows form and ingress around all nuclei in these embryos as in wild-type. These furrow canals contract during late cellularization when the basal surface of the cell is normally closed-off from the yolk mass. Furrow canals are discrete tear-shaped compartments. Furrow tips broaden to form distinct furrow canal structures in these embryos.

When homozygous mutant germ-line clones are made, mutant embryos disintegrate, as cell delaminate owing to an inability to form adherens junctions.

Scer\GAL4arm.PU-driven expression of sggScer\UAS.T:Myr-Src64B,T:Ivir\HA1 (but not sggScer\UAS.T:Ivir\HA1) can mildly rescue cuticle integrity in arm043A01 maternally and zygotically mutant arm043A01 embryos.

One copy of SCARΔ37 partially suppresses the cuticle defects seen in arm043A01 mutant embryos.

One copy of Arp3EP3640 partially suppresses the cuticle defects seen in arm043A01 mutant embryos.

Embryos double mutant for arm043A01 both maternally and zygotically and l(2)gl4 zygotically display abnormal embryonic cuticle.

Expression of nulloScer\UAS.T:Ivir\HA1, under the regulation of Scer\GAL4mat.αTub67C.T:Hsim\VP16 in an arm043A01 mutant, results in cells that are unable to flatten or constrict their apical surfaces as in nulloScer\UAS.T:Ivir\HA1 (Scer\GAL4mat.αTub67C.T:Hsim\VP16) single mutants.

Comments

The adhesion defect of arm043A01 is not rescued by expression of armΔ.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4arm.PS. The adhesion defect of arm043A01 is rescued by expression of armS10.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4arm.PS.
