Detailed Description

Mutant larvae show reduced encapsulation (10%) of eggs when parasitised by the avirulent L. boulardi wasp strain G486. Plasmatocytes fully cover the egg, while lamellocytes partially cover it.

Homozygous larvae show a reduced rate of encapsulation (27%) of eggs of the avirulent wasp strain L. boulardi G486 compared to the rate seen in wild-type larvae. By 20 hours post-parasitisation, the wasp eggs are completely encapsulated by plasmatocytes, which are properly spread around the wasp egg chorion, as occurs in wild type. However, at 38-40 hours post-parasitisation, eggs from mutant larvae have fewer lamellocytes adhered to them than those from wild-type larvae. 40% of the hemocytes bled from these mutant larvae have abnormal elongated extensions.

Isolated plasmatocytes from homozygous larvae show a reduced ability to phagocytose E. coli and S. aureus in a phagocytosis assay.

ZirBG00267/+ ; Rac2Δ/+ double heterozygous larvae have a reduced rate of encapsulation (42%) of eggs of the avirulent wasp strain L. boulardi G486 compared to the rate seen in either single heterozygote.

ZirBG00267/+ ; Cdc422/+ double heterozygous larvae have a reduced rate of encapsulation (37%) of eggs of the avirulent wasp strain L. boulardi G486 compared to the rate seen in either single heterozygote.

Expression of either Cdc42V12.Scer\UAS or Rac2Scer\UAS.cARa under the control of Scer\GAL4He.PZ rescues the reduced rate of encapsulation of eggs of the avirulent wasp strain L. boulardi G486 that is seen in ZirBG00267 larvae.

Expression of either Cdc42V12.Scer\UAS or Rac2Scer\UAS.cARa under the control of Scer\GAL4He.PZ rescues the reduced phagocytosis index against E. coli and S. aureus seen in isolated plasmatocytes from homozygous ZirBG00267 larvae.
