antennal glomerulus & neuron, with Scer\GAL4GH146

olfactory sensory organ & neuron & dendrite, with Scer\GAL4GH146

Detailed Description
Statement
Reference

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4acj6-PG63 in a acj6PG63/Y background is able to induce a fenchone odor response, indicating activation of Or85e.

When acj61,4.Scer\UAS is driven by Scer\GAL4GH146 in the longitudinal projection neurons (lPN) three phenotypes are seen. Reduces innervation of appropriate glomeruli, non-specific accumulation and specific mistargeting to inappropriate glomeruli. lPN misexpressing acj61,4.Scer\UAS still innervate the same 10 lateral landmark glomeruli (DA1, DL3, DA2, DM5, DM2, DM1, VA7m,VA5, VA4 and VC2) as wild-type, though they occasionally innervate some glomeruli more weakly or partially. Secondly they exhibit diffuse dendritic projections that span part of the lateral side of the antennal lobe with associated glomerular abnormalities. Thirdly mutant lPNs also mistarget their dendrites to well developed landmark glomeruli that are normal reserved for anterodorsal projection neurons (adPNs). This mistargeting often leads to only partial occupation of these glomeruli. It appears to be a specific effect, since glomeruli with mistargeted dendrites are frequently seen surrounded by uninnervated glomeruli.

Expression of acj61,4.Scer\UAS under the regulation of Scer\GAL4Cha.7.4 results in third instar larval or late pupal stage lethality.

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4sca-537.4 results in a failure of motor axons of the ISNb fascicle to defasciculate from the ISN fascicle, resulting in defective innervation of the ventrolateral muscle field in approximately 65% of embryonic hemisegments. In approximately 15% of hemisegments, the ISNb motor axons stop at the correct location of their target muscles, but do not branch out to innervate the ventral muscle groups. The remaining 20% of hemisegments show a wild-type muscle innervation pattern.

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4GH146 in homozygous ems3 adPN neuroblast clones (induced in the early larva and analysed in the adult) significantly rescues the innervation defects of the VA1lm glomerulus, but does not significantly rescue the innervation defects of the VA3 and VM2 glomeruli which are seen in single mutant homozygous ems3 adPN neuroblast clones.

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4acj6-PG63 rescues the response of the sensilla pb1A neuron to ethyl acetate, isoamyl acetate, 4-methyl phenol, benzaldehyde and fenchone.

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4acj6-PG63 in a acj6PG63/Y background is able to induce a fenchone odor response, indicating activation of Or85e.

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4acj6-PG63 in a acj6PG63/Y background suppresses the (pb2C, 0) type of sensilla normally found in acj6PG63 mutants.

Expression of acj61,4.Scer\UASunder the control of Scer\GAL4acj6-PG63 in a acj6PG63/Y background induces two unexpected types of sensilla. One type contains a normal pb2B cell and a novel ORN that responds to 3-octanol but to no other odorants. This ORN is termed here pb2D. This (pb2D, pb2B) sensillum represents 25 and 21% of the sensilla containing pb2A or pb2B neurons in flies expressing acj61,4.Scer\UAS and acj61,3,4.Scer\UAS, respectively. The second type of sensilla, representing 45% of the pb2 sensilla in acj61,4.Scer\UAS expressing flies, respectively, contains a normal pb2B cell and a novel ORN that responds to both 3-octanol and fenchone and is termed her pb2E.

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4acj6-PG63 in a acj6PG63/Y background, fully rescues the sensilla pb1 phenotype, yielding (pb1A, pb1B) sensilla that respond as expected to all odorants tested (ethyl acetate, isoamyl acetate, 4-methyl phenol, benzaldehyde, 3-octanol and 4-propyl phenol).

Expression of acj61,4.Scer\UAS under the control of Scer\GAL4acj6-PG63 in a acj6PG63/Y background rescues the pb3 sensilla.
