Description

The Mer coding region lacking amino acids 171 to 177 in the conserved N-terminal region and tagged at the N-terminal end with Tag:MYC, is expressed under the control of UASt regulatory sequences.

Expression of MerΔBB.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ap.PU in cells of the wing disc proper has no significant effect on the morphology or proliferation of the overlying peripodial membrane cells in third instar larvae. The wings of the adult flies however curl downwards due to the overgrowth of the dorsal surface.

Flies carrying one copy of MerΔBB.Scer\UAS.T:Hsap\MYC expressed under the control of one copy of Scer\GAL4en-e16E have a moderately overgrown posterior wing compartment with no disruption in venation. Flies carrying two copies of MerΔBB.Scer\UAS.T:Hsap\MYC expressed under the control of two copies of Scer\GAL4en-e16E show an enlargement of the posterior wing compartment with disruptions in venation, particularly along vein 5 and the posterior crossvein. Disruptions include formation of ectopic vein material. The wings are also held out from the body axis.

Flies expressing MerΔBB.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E show expansion of the wing blade. This phenotype is enhanced if the flies are also heterozygous for Mer4, and is suppressed if the flies are also carrying Mer+t36. Flies expressing MerΔBB.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ap-md52 have wings heldout at an average of 45o from the body axis. This phenotype is greatly suppressed by Mer1-600.Scer\UAS.T:Hsap\MYC and slightly suppressed by MerScer\UAS.T:Hsap\MYC.

Flies carrying one copy of MerΔBB.Scer\UAS.T:Hsap\MYC expressed under the control of one copy of Scer\GAL4en-e16E and also carrying one copy of Df(2R)Px2 or bs364 show wing blistering. The overgrowth of the posterior compartment seen in flies carrying one copy of MerΔBB.Scer\UAS.T:Hsap\MYC expressed under the control of one copy of Scer\GAL4en-e16E is enhanced by sbb94, sbb256, sbb270, E(Mer)IIb187, E(Mer)IIb209, bs242, net383 or E(Mer)IIIa320. The wing phenotype seen in flies carrying one copy of MerΔBB.Scer\UAS.T:Hsap\MYC expressed under the control of one copy of Scer\GAL4ap-md52 is modified by bs242, sbb256, net383, E(Mer)IIb187 or E(Mer)IIIa239. Expression of MerΔBB.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ap-md52 in combination with either sbbunspecified or bsunspecified results in a significant blistered wing phenotype.
