Description

P{lacW} insertion in an intron 1917bp before the first exon common to mRNAs encoding shot isoforms A and B.

Insertion of a P{lacW} element into the intron at the 5' end of the gene that separates the alternate starts from the shared exons.

shotk03010/Df(2R)BSC361 transheterozygotes do not display visible microtubule distribution defects in third instar larval adipocytes.

shotk03010 homozygous and shotk03010/shotΔEGC transheterozygous embryos show a smaller dorsal hole width during dorsal closure, as shown by a significant increase in the length:width ratio, and the leading edge of their dorsal-most epithelial cells do not display significant changes in cell size, but present a significant decrease in the number of filopodia, present frequent long apical protrusions and present frequent abnormally long, bent and protruding microtubules, as compared to controls.

When homozygous germ-line clones are made, no defect is seen in oocyte determination.

The lateral chordotonal sensory axons arrest at variable stages before reaching the central nervous system (CNS); 27% extend towards the intersegmental nerve (ISN) and stall or fail to reach the ISN, 55% travel a significant distance within the ISN, 18% reach the CNS but do not penetrate it, and none of the axons enter the CNS and form arborisations.

In the dorsal cluster of embryonic dendrites of shot30/shotk03010 embryos, the number of anterior lateral branches is reduced from about 10 to 4 per hemisegment, whereas more branches extend dorsally and appear to contact each other more extensively.

Germ band retraction and head involution fail in approximately 15% of homozygous embryos.

Lethality is reverted by excision of the P{lacW} element.

The site of the P{lacW} insertion in shotk03010 is identical to the site of the P{lacW} insertion in shotk03405.
