The posterior compartment of the pupal wing undergoes excess pigmentation (showing black rather than brown pigmentation) in the presence of dopamine in an in vitro melanisation assay in animals expressing yScer\UAS.cCa under the control of Scer\GAL4en-e16E.
When yScer\UAS.cCa is driven by Scer\GAL4pnr-MD237, pigmentation in the thorax is slightly darkened compared to wild-type flies. The colour intensity and the anterior posterior width of the abdominal pigmentation is mildly increased. The lateral width of the pigment along the dorsal midline also increases. However in many cells, such as those anterior to the pigment stripe, no black pigemtn is induced.
When expressed under the influence of Scer\GAL4hs.PB, heat shocks given at or after the second half of the third larval period are able to produce y+ rescue. When expressed via Scer\GAL4Dll-md23 the tibia, basitarsus and tarsal segments and a small region at the trochanter-femur joint are pigmented, as well as along the dorsoventral boundary in the wing and in the distal antennal segments. When expressed via Scer\GAL4cad-em459 the analia are pigmented. When expressed via Scer\GAL4pnr-MD237 a dorsal band of pigment extends the length of the body from the occipital head region to the end of the abdomen but excluding the terminalia. In the thorax the piment extends from the midline to a longitudinal straight line defined by the dorsocentral bristles. When expressed via Scer\GAL4ost-em462 pigment is expressed in a band adjacent to the pnr domain revealed by Scer\GAL4pnr-MD237.
Scer\GAL4pnr-MD237, yUAS.cCa has abnormal body color phenotype, enhanceable by e1
yUAS.cCa/Scer\GAL4pnr-MD237 is a suppressor | partially of abnormal body color phenotype of Scer\GAL4pnr-MD237, eUAS.cWa
Scer\GAL4pnr-MD237, tUAS.cTa, yUAS.cCa has abnormal body color phenotype
Scer\GAL4pnr-MD237, tUAS.cTa, yUAS.cCa has adult cuticle phenotype
Co-expression of both yScer\UAS.cCa and tScer\UAS.cTa under the control of Scer\GAL4pnr-MD237 produces dramatic black pigmentation in the Scer\GAL4pnr-MD237 expression domain of the adult abdomen in males.
When yScer\UAS.cCa is driven by Scer\GAL4pnr-MD237 in a e1 background, a dramatic increase in black pigment in the thorax and in the abdomen. When yScer\UAS.cCa and eScer\UAS.cWa are driven together by Scer\GAL4pnr-MD237 induces the formation of more black melanin than eScer\UAS.cWa alone, though the wild-type level of pigmentation is not restored.
yUAS.cCa/Scer\GAL4dsx-GAL4 rescues y1
Scer\GAL4dsx.KI/yUAS.cCa rescues y1
yUAS.cCa/Scer\GAL4c81 rescues y1
yUAS.cCa/Scer\GAL4Act5C.PI rescues y1
yUAS.cCa, Scer\GAL80nSyb.PS, Scer\GAL4dsx-GAL4 partially rescues y1
yUAS.cCa/Scer\GAL4y.PG partially rescues y1
yUAS.cCa/Scer\GAL4pnr-MD237 partially rescues yunspecified
Scer\GAL4fru.P1.D/yUAS.cCa fails to rescue y1
The expression of yUAS.cCa under the control of Scer\GAL4y.PG suppresses the yellow body color but not the male mating defects of y1 mutants.
The expression of yUAS.cCa under the control of Scer\GAL4dsx-GAL4 alone or under the combined control of Scer\GAL4dsx-GAL4 and Scer\GAL80nSyb.PS fully and partially rescues, respectively, the male mating defects of y1 mutants.
The expression of yUAS.cCa under the control of Scer\GAL4dsx.KI rescues both the body color and male mating defects of y1 mutants.
Expression of yScer\UAS.cCa under the control of Scer\GAL4pnr-MD237 in yunspecified males rescues the y mutant phenotype only in the overlapping domain of Scer\GAL4pnr-MD237 expression and the endogenous brown pigmentation.
The reduced wing extension index (the percentage of each courtship ritual during which a male's wing is extended and vibrating) seen in y1 males is rescued to roughly that of wild-type males by expression of yScer\UAS.cCa under the control of Scer\GAL4Act5C.PI or Scer\GAL4c81.
