Description

The loxP cassette containing the Avic\GFPGMR.PS marker has been excised. This results in a fru locus in which the endogenous fru P1 promoter drives expression of GAL4 instead of the fru[M] isoform. A transcriptional stop signal and a single loxP site remain downstream of the GAL4 open reading frame.

Drives expression in most fruitless expressing neurons in the adult, including neurons of the male and female reproductive systems, and a small subset of olfactory receptor neurons.

Expression driven by Scer\GAL4fru.P1.D was restricted via a transgenic cell class-lineage intersection (CLIn) system to the following subset of sexually dimorphic fruitless neurons of specific Type II neuroblast lineages using Klac\KDRstg14; 27-39 male and 15-27 female pMP-a neurons - part of the DM1 lineage, 19-27 male and 7-11 female pMP-b neurons - part of the DM2 lineage, 3 male and 1-3 female pMP-c neurons - part of the DM3 lineage, 46-50 male and 4-10 female pMP-e neurons - part of the DM4 lineage, 10-14 pIP-g neurons - part of the DM5 lineage, 2 pIP-j neurons - part of the DM5 lineage, 24-26 male and 12-24 female pMP-f neurons - part of the DM6 lineage, a single pIP-h neuron - part of the DM6 lineage, 20-34 male and 16-24 female pIP-e neurons - part of the DL1 lineage, and 17-27 male and 16-24 female pIP-a neurons - part of the DL2 lineage. All of these neurons show sexually dimorphic differences in their projections. In total, ~19% (~396/2083) male and ~8.3% (~164/1980) female Scer\GAL4fru.P1.D neurons intersect with Klac\KDRstg14.

Scer\GAL4fru.P1.D drives expression in Ilp7 positive postembryonic neurons.

Scer\GAL4fru.P1.D labels the olfactory receptor neurons Ir84a, Or67d and Or47b. It is expressed in the antennal lobe glomeruli VL2 anterior compartment, DA1 and VA1 ventral compartment.

Scer\GAL4fru.P1.D drives expression in neurons of the female and male gonads. Scer\GAL4fru.P1.D labels 8 neurons on the female lateral oviduct and uterus and 2 neurons in the male ejaculatory duct. All these neurons co-express Avic\GFPEGFP.ppk.1.

Scer\GAL4fru.P1.D drives expression in 2-3 Avic\GFPEGFP.ppk.1 positive fruitless expressing neurons either side of the uterus.

Inactivation of a subset of Scer\GAL4fru.P1.D cells (using Ctet\tetXTNT-LC.FRT.stop.UAS and Scer\FLP1402) in males results in death within 4 d of eclosion. Inactivation of a different subset of Scer\GAL4fru.P1.D cells (using Ctet\tetXTNT-LC.FRT.stop.UAS and Scer\FLP1335) in males significantly reduces courtship index, frequency of successful matings and copulation duration and significantly increases copulation latency, compared to controls; when inactivation is restricted to a smaller subset of cells (using Scer\GAL80tsh-GAL80), the frequency of successful matings is partially rescued, but still significantly lower than controls, copulation latency is no longer significantly different from controls, but copulation duration is still significantly lower than controls.

Activation of Scer\GAL4fru.P1.D neurons triggers wing extension behavior; depending on the combination of UAS and light used, different numbers of flies show this behavior: 0% with blue light activation of Crei\ChR2Scer\UAS.cSa, Crei\ChR2H134R.Scer\UAS.2A.T:Avic\GFP-EYFP or Crei\ChR2C128T.Scer\UAS.T:Avic\GFP-YFP; 0% with red or green light activating Crei\ChR1[C1V1.T-T.Scer\UAS. T:Avic \GFP-EYFP]; 80% (red) or 100% (green) light activating Crei\ChR1ReaChR.Scer\UAS.T:Crei\ChR2,T:Avic\GFP-mCitrine.
