Description

Construct: Residues 1-455 of tor13D, including the extracellular and transmembrane domain, fused to the entire ORF of tub.

First 455 amino acids of tor, containing the signal peptide, extracellular domain with a single amino acid replacement and transmembrane peptide, are fused to amino acids 2-462 of tub.

Detailed Description
Statement
Reference

When injected into embryos from snk1/snk2 mothers, filzkorper and ventral denticles show 84% rescue. tor::tubWT and tor::tubt4021t both induce filzkorper and denticle belts, but tor::tubt4021t has a much higher activity.

tor::tubt4021t is a non-suppressor of embryo | ventral phenotype of pll7

Additional Comments
Genetic Interactions
Statement
Reference

When tor::tubt4021t is injected into embryos laid by snkunspecified mothers, significant rescue of the dorsalised phenotype is seen. Some ventrolateral pattern elements are seen, such as ventral denticle belts and Filzkorper, which are never seen in Myd88kra-1 mutant embryos.

Injection of RNA into wild type, gd and tub null embryos rescues ventral fate, filzkorper and ventral denticle belts are evident. Injection into pll7 embryos effects no ventral rescue.

Carried in plasmid "tor4021tub" and used in transient expression assays in embryos.

Transcribed in vitro and injected into Myd88kra-1 or snkunspecified embryos to study interaction.

Ventralising gain of function allele of tub.

Carried in plasmid "tor4021-tub", used to generate RNA for microinjection studies.
