Description

A genomic per fragment containing 4.2kb 5' flanking sequences and part of the coding region (encoding the N-terminal 638 amino acids) is fused in frame to lacZ.

4kb fragment of per 5' flanking sequences upstream of the transcription start site, the same extent as in per+t13.2, and genomic sequences up to that encoding approximately the first 630 amino acids, within the fifth exon, are fused to lacZ.

Expression was first seen in 12-14hr old first instar larvae. Expression was detected by X-gal histochemistry in DN2, but not DN1, in L2 larvae. Throughout larval development, expression is seen in cells which do not cycle per found in the ventral region of the brain.

The phase of the morning activity peak of flies carrying perSG.T:Ecol\lacZ in a per+ background is similar to that of wild-type controls, while the phase of the evening activity peak is significantly different, being earlier than wild-type controls, when flies are kept under 12 hour light/12 hour dark cycling conditions. The free-running locomotor activity periods of flies carrying perSG.T:Ecol\lacZ kept under DD conditions are significantly shorter than wild-type controls, and only 70-80% of the flies are rhythmic under these conditions.

Has little or no effect on circadian locomotor activity rhythms in a per+ background.
