No phenotypes are observed in ds05142 mutant DL1 olfactory projection neurons.

Neuroepithelial overproliferation results in grossly distorted brains with large optic lobes in ds05142 late third instar larvae.

dsUAO71/ds05142 mutant wings display abnormal morphology. The region between wing veins L4 and L5 of dsUAO71/ds05142 mutant wings retains wild-type hair polarity, but has primarily anterior-posterior ridges.

ds05142 larvae show mild wing disc overgrowth.

ds05142 adults have wings with widespread planar cell polarity defects and that are partially lacking the crossveins.

ds05142/ds05142 flies have severe planar polarity defects in their wings.

Occasionally in ds38k/ds05142 escapers (approximately 2% of heminota), the wing is replaced by a winglet and, posterior to it, a lateral protuberance that looks like an scutum and scutellum. The winglet is composed of proximal anterior structures arranged in a mirror-image duplication. The smallest examples of these winglets consist of a mirror image duplication of anterior hinge structures - the tegula and humeral sclerite. Larger examples also have a rudimentary wing blade consisting of a mirror image duplication of anterior structures - the costa and anterior wing margin. ds05142/ds1 flies have legs with reduced segment size and fusion of the tarsal segments and partial elimination of the tarsal joints.

Escapers or pharate adults have a strong ds mutant phenotype.

Expression of dsScer\UAS.cTa or dsΔICD.Scer\UAS, under the control of Scer\GAL4αTub84B.PL, rescues the planar cell polarity defects of ds05142 wings in all areas except the most proximal regions of the wing blade. Expression of either transgene driven by Scer\GAL4ds-gal4 partially rescues planar cell polarity in the proximal wing but does not fully rescue the loss of the crossveins.

The planar polarity phenotype of ds05142/ds05142 wings is not enhanced by fjd1/fjd1.

dsh3/+ enhances the severity of leg phenotypes in ds05142/ds1 animals: The length of the segments is reduced and most of the tarsal joints very reduced or completely absent.

Comments

Uniform dsScer\UAS.cTa expression using Scer\GAL4αTub84B.PP rescues the planar cell polarity defects of ds05142/ds05142 flies in the distal region between L3 and L4, but not in the proximal region just posterior to L1. Planar cell polarity defects in the posterior compartment of ftScer\UAS.cMa; Scer\GAL4en-e16E flies are weakly suppressed by ds05142/ds05142.
