The mir-125 seed sequence (TCCCTGAGACCCTAA) has been deleted from the progenitor construct.

The P{W8,let-7-CΔmiR-125} construct can rescue the male fertility and climbing defects of Df(2L)let-7-CKO1/Df(2L)let-7-CGKI adults. However, the rescued adults still show severely reduced locomotion and show partial defects in flight. Df(2L)let-7-CKO1/Df(2L)let-7-CGKI females rescued by P{W8,let-7-CΔmiR-125} show normal fertility.

Df(2L)let-7-CKO1/Df(2L)let-7-CGKI adults rescued by the P{W8,let-7-CΔmiR-125} construct no longer show abnormal persistence of the dorsal internal oblique muscles (these muscles decay during post-eclosion maturation in wild-type animals) and no longer show defects in the dorsal muscles and their neuromuscular junctions.
