Interaction in vitro; bait produced as a recombinant fusion protein in bacterial system; prey derived from wild-type embryonic extract.

Hundreds of potential jar-interacting proteins identified by mass spectrometry were compared to proteins pulled down by another myosin, didum; only those proteins identified by five times as many unique peptides in the jar pulldown, compared to the didum pulldown, were considered. To further refine the list of potential jar-interacting proteins, the ratio of unique peptides to predicted molecular weight (kDa) was calculated and those with a ratio greater than that for mira protein (a known jar interacting protein) were considered further.
