ECDYSTEROID KINASE-LIKE -
Members of the arthropod-specific ecdysteroid kinase-like (EcKL) gene family are hypothesised to encode detoxicative kinases responsible for detoxification-by-phosphorylation. They are predicted to conjugate phosphate to secondary alcohols, using ATP as a phosphodonor. However, so far only one EcKL enzyme (ecdysteroid 22-kinase from Bombyx mori) has been characterised, phosphorylating ecdysteroids (insect growth and moulting hormones) at C-22, to form physiologically inactive ecdysteroid 22-phosphates. Mutation in sequenced strain: insertion of transposon; an insertion allele segregating in wild-type strains; see CHKov1[Doc1420].

The Doc transposable element insertion is correlated with pesticide resistance (FBrf0188316) and resistance to sigma virus (FBrf0216506); may be due to effect upon CHKov2 (FBrf0216506), rather than a function of atypical CHKov1 transcripts.

Atypical transcripts containing Doc sequences have been described (FBrf0188316) and were included in the FlyBase gene model annotation prior to release 5.53.

          l(3)rJ880 may correspond to CHKov1: the P{PZ}l(3)rJ880rJ880 insertion maps within the transcription unit.

          Insertion of a Doc element into CHKov1 (Doc{}CHKov1Doc1420) increases resistance to Sigma virus compared to control flies lacking the insertion. The rearranged Dp(3;3)CHKov chromosome, which is derived from the Doc{}CHKov1Doc1420 chromosome, shows a further increase in resistance to Sigma virus compared to the Doc{}CHKov1Doc1420 progenitor. Since the rearrangement amplifies the region coding for CHKov1 containing the Doc insertion, the simplest explanation for the increased resistance to Sigma of the Dp(3;3)CHKov chromosome is that the amplification of the region coding for CHKov1 containing Doc increases the resistance. However, no evidence has been found for the expression of CHKov1 changing in Dp(3;3)CHKov relative to Doc{}CHKov1Doc1420, while the Dp(3;3)CHKov rearrangement is associated with a 6- to 9-fold increase in expression of the related CHKov2 gene. Thus it is possible that both CHKov1 and CHKov2 play a role in resistance to Sigma virus (and thus that the locus previously referred to as "ref(3)D" corresponds to CHKov1 and CHKov2).
