prt transcript is detected in the adult head, but not in the adult body. There is also some expression in the second and third instar larval stages.
In the third larval instar, prt protein is expressed in one to two cell bodies per hemisegment in the ventral nerve cord. These cells project medially into the neuropil in a complex pattern. Strong expression in the adult head is confined to a subset of Kenyons cells and in the calyx, vertical and medial lobes and pedunculus. prt protein was also detected in one bilateral extrinsic neuron that projects to the vertical and medial lobes of the mushroom bodies. In the adult, prt protein is expressed in all of the lobes of the mushroom bodies and calyx. Expression in the pedunculus is not uniform, showing weak labelling in the core. Apart from the mushroom bodies, prt protein is detected in 56 cell bodies in the brain. These include cells near the mushroom bodies, two to three cells in the subesophageal ganglion which project medially towards the esophogeal foramen, in the central body (fan-shaped and ellipsoid bodies) and two bilaterally symmetric clusters of two to three cells near the medial aspect of the optic lobe which project toward the medulla. In the thoracico-abdominal ganglion, prt protein is detected in three clusters of two to four cells each are found near the ventral midline.
