Detailed Description

MhcR249Q/+, Mhc10/Mhc10 individuals do nor show any obvious defects in DLM fibers.

Both Mhc+t24.1 hetero- and homo-zygosity in a Mhc10 background is viable and adults can fly. In the Mhc+t24.1, Mhc10 double homozygosity condition, pupal and adult indirect flight muscles show regularly packed myofibrils, with six thin filaments around each thick filament and well-formed sarcomeres.

Flies expressing Mhc+t12.4 are able to jump 1.5-fold farther than Mhc10 mutants.

Relaxed tension of tergal depressor of the trochanter (TDT) muscle expressing Mhc+t12.4 is very low, especially compared to the indirect flight muscles. Net active tension is approximately 30-fold higher than passive tension in Mhc+t12.4 flies, whereas all myosins tested in the indirect flight muscles exhibit passive tension levels almost equal to net active tension. TDT expressing Mhc+t12.4 exhibit a very steep tension-pCa curve.

Comments

Mhc10 mutants expressing Mhc+t12.4 display normal jumping ability and ultrastructure.

The addition of Mhc+t24.1 rescues flight levels in a Mhc1 heterozygous background to wild-type levels.
