Detailed Description

Lmx1aΔ1 homozygous and Lmx1aΔ1/Df(3L)ED4475, Lmx1aΔ1/Df(3L)ED4483 and Lmx1aΔ1/Df(3L)BSC380 transheterozygous females show complete or near-complete loss of ovariole material/ovaries; Lmx1aΔ1 homozygous females do not lay any eggs. In limited genetic backgrounds, including a non-backcross Lmx1aΔ1/Tm3 balanced stock, residual ovary tissue can be seen in newly eclosed virgin Lmx1aΔ1 homozygous females but without morphologically recognizable terminal filaments.

Early at the larval-pupal transition, Lmx1aΔ1 ovaries show partial evidence of terminal filament morphogenesis but these do not form the collection of fully formed, linear stacks; by 24h/48h after puparium formation, terminal filaments are absent, ovaries show no sign of growth and elongation, and primordial ovarioles do not form.

Comments

Expressing Lmx1aUAS.ORF.GW.Tag:HA under teh control of Scer\GAL4GMR42B09 significantly restores overall ovary morphogenesis, including sheath development, TF formation and egg laying in Lmx1aΔ1 homozygous females. Ovary morphology is also partially rescued if this expression is only induced from mid-L3 but not from 2h after pupariation (using Gal80[ts] to temporally control the expression).
