Under constant darkness, individuals expressing mir-210UAS.cCa under the control of Scer\GAL4tim.UAS show prevalent circadian arrhythmicity; the remaining rhythmic individuals show a lengthened period, but with 5 h phase delay, as compared to controls. Under 12h:12h light:dark cycles, individuals lose their ability to anticipate the lights-ON transition (as shown by Morning Index and Morning Anticipation values) and delay the evening activity onset, as compared to controls. The arrhythmicity is not observed when expression is only induced during development and is less severe when the expression is only induced during adulthood (using Gal80[ts] to temporally control the expression); individuals stitl fail to anticipate Lights-ON. Expression ristricted to development also leads to a decreased optomotor response.
The expression of mir-210UAS.cCa under the control of Scer\GAL4tim.UAS abolishes the daily changes in s-LNv neuron axon termini (i.e. shift from defasciculated state at ZT3 to fasciculated state at ZT15) observed in controls, thus remaining at the fasciculated state; this phenotype is not observed when the expression is restricted to either development or adulthood.
The expression of mir-210UAS.cCa under the control of Scer\GAL4tim.UAS, Scer\GAL4dimm-929 or Scer\GAL41118, but not Scer\GAL4P2.4.Pdf, leads to I-LNv (Pdf-positive) neurons to exhibit aberrant arborizations in the optic lobe, as well as an altered cell body shape, as compared to controls; Scer\GAL4tim.UAS-driven expression still leads to aberrant I-LNv arborizations if restricted to development, but not to adulthood.
Under constant darkness, individuals expressing mir-210UAS.cCa under the control of Scer\GAL4cry.PE, but not Scer\GAL41118, Scer\GAL4P2.4.Pdf, Scer\GAL4mir-210-KO (the intrinsic driver of mir-210KO) or Scer\GAL4dimm-929 leads to prevalent circadian arrhythmicity. Under 12h:12h light:dark cycles, individuals expressing mir-210UAS.cCa under the control of either Scer\GAL4cry.PE, Scer\GAL41118 or Scer\GAL4P2.4.Pdf, but nor Scer\GAL4mir-210-KO or Scer\GAL4dimm-929, results in delayed evening activity onset, as compared to controls.
Scer\GAL4mir-210-KO/mir-210UAS.cCa partially rescues mir-210KO
Scer\GAL4mir-210-KO, mir-210UAS.cCa, Scer\GAL80Pdf.PS partially rescues mir-210KO
Scer\GAL80cry.PS, Scer\GAL4mir-210-KO, mir-210UAS.cCa partially rescues mir-210KO
Expressing mir-210UAS.cCa under the control of Scer\GAL4mir-210-KO (the intrinsic driver of mir-210KO) mostly rescues the circadian defects of mir-210KO mutants under either constant darkness or 12h:12h light:dark cycles (LD). although the advanced evening onset under LD is only partially rescued.
Expression under the additional control of Scer\GAL80cry.PS rescues the advanced evening activity onset but not activity phase delay of mir-210KO.
Expression under the additional control of Scer\GAL80Pdf.PS does not rescues the advanced evening activity onset nor the activity phase delay of mir-210KO mutants.
