Description

Sequence encoding a GAL4 driver and a w+* marker are inserted into the Tmc locus near the translation initiation codon of Tmc isoform PD, resulting in about 7,000 bp of the Tmc gene being replaced by the reporter construct and more than 1,000 amino acids (about one-half of the coding sequence) of Tmc being deleted.

TmcGal4/Tmc1 mutant adult females have a significant increase in preference for softer 100 mM sucrose-containing, but not plain, agarose for egg laying, compared to controls; TmcGal4 homozygous, but not heterozygous, adult females also have a significant increase in preference for softer 100 mM sucrose-containing agarose for egg laying, compared to controls; TmcGal4/Tmc1 and TmcGal4 homozygous adult females have no difference in preference for 100 mM sucrose-containing (rather than plain) agarose for egg laying, compared to controls.

TmcGal4 homozygotes as well as TmcGal4/Df(3L)BSC576 mutants display significant larval locomotion defects: reduced crawling speeds, increased head cast frequencies and enhanced backward locomotion.

TmcGal4 homozygote mutants do not display any defects in either the dendrite morphology of class I dopaminergic neurons or in axon projections of the Tmc-positive neurons in the ventral nerve cord.

Expression of Mmus\Tmc1Scer\UAS.cGa or Mmus\Tmc2Scer\UAS.cGa - individually as well as together - under the control of Scer\GAL4Tmc-Gal4 rescues (or partially rescues in the case of Mmus\Tmc2Scer\UAS.cGa single expression) the larval locomotion defects characteristic for TmcGal4 homozygotes (increased head curl frequency, tendency to backward locomotion).
