Homozygous, hemizygous and trans-heterozygous (with exp11) mutant embryos exhibit bubble-like cysts with an enlarged diameter in unicellular (lateral trunk, ganglionic branch) and intracellular (lateral ganglionic branch) tracheal branches, but not in multicellular branches (dorsal trunk) or intracellular fusion branches. Bubbles are also seen in the dorsal branch and visceral branch, but to a lesser extent. Despite this, the tracheal network exhibits normal branching patterns. Tracheal cell number is also unaffected in exp135 mutants.

Tracheal cysts in exp135 embryos are modified apical extracellular lumen in unicellular branches, and intracellular vacuole-like compartments in intracellular branches.

Defective accumulation of a subset of luminal components is observed in unicellular and intracellular (but not multicellular) tracheal branches where cysts form in exp135 mutants.

Scer\GAL4btl.PS-mediated expression of Rho1V14.Scer\UAS enhances, whereas expression of Cdc42V12.Scer\UAS or Rac1N17.Scer\UAS or Rho1N19.Scer\UAS partially suppresses the tracheal cyst phenotype of exp135 embryos. Scer\GAL4btl.PS-mediated expression of Cdc42N17.Scer\UAS has no obvious effect on this phenotype.
