Driving Or83cScer\UAS.cRa with Scer\GAL4Or23a.PC in Or83cMB11142 mutant background confers farnesol sensitivity to ai2b neurons.

Responses to farnesol are moderately reduced in Scer\GAL4Or67d-GAL4-2>Or83cScer\UAS.cRa transgenic flies in an ATP8Bf05203 mutant background compared with transgenic expression in a wild-type genetic background.

Flies homozygous for Or67dGAL4-2 and expressing Scer\GAL4Or67d-GAL4-2>Or83cScer\UAS.cRa show restored spontaneous activity in at1 neurons, but to a distinct higher average firing rate. The mutant neurons expressing Or83cScer\UAS.cRa remain insensitive to the male-specific pheromone 11-cis-vaccenyl acetate (cVA), similar to Or67dGAL4-2 mutants.

A single odorant out a panel of approximately 150 tested volatile compounds, farnesol elicited robust activity in the mutant at1 neurons expressing Scer\GAL4Or67d-GAL4-2>Or83cScer\UAS.cRa. Importantly, farnesol does not activate wild-type at1 neurons.

Driving Or83cScer\UAS.cRa with Scer\GAL4Orco.2.642.T:Hsim\VP22 restores farnesol sensitivity to ai2b neurons in Orco2 mutants.

Driving Or83cScer\UAS.cRa with Scer\GAL4Snmp1.PR restores farnesol sensitivity to ai2b neurons in Snmp1Z0429 mutants.

Or83cScer\UAS.cRa fails to confer farnesol sensitivity when mis-expressed under the control of Scer\GAL4Or22a.8197 in a basiconic neuron context in a Df(2L)dp-79b/Dp(2;2)dppd21 ("Δhalo") genetic background.
