plumΔ1 homozygotes show adult mushroom body defects:

late-born β/β'-axons over-extend beyond the midline in about half brains and often the β/β'-lobes from both hemispheres seem to fuse; γ-neurons show severe pruning defects.

plumV11X homozygous β-axons only over-extend beyond the midline if in a plumV11X homozygous background, but not in a heterozygous background.

plumΔ1 mushroom body gamma neuron clones frequently show pruning defects.

The transverse nerve makes inappropriate connections at the neuromuscular junction in plumΔ1 larvae.

Homozygous mushroom body neuroblast clones show a severe pruning defect at the pupal stage.

8% of homozygous single cell mushroom body neuroblast clones in an otherwise heterozygous background fail to prune their branches, retaining their larval-specific dorsal axon branches into the adult stage. The fraction of unpruned single cell clones is increased if the homozygous plumΔ1 cells are also expressing plumKK103873 under the control of Scer\GAL4Tab2-201Y.

The frequency of improper, ectopic projections at the larval neuromuscular junction is increased compared to wild-type controls, with the majority of the ectopic connections being made by the transverse nerve.

Expression of plumScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4Tab2-201Y fully rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

Expression of plumΔIg1-3.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4Tab2-201Y rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

Expression of plumΔIg1-4.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4Tab2-201Y partially rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

Expression of plumΔIg4.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4Tab2-201Y shows very little ability to rescue the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones (rescue seen in 1 out of 16 cases).

Expression of plumΔCyt.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4Tab2-201Y rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

plumT:Avic\GFP,T:Ivir\HA1 rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

plumΔCyt.T:Ivir\HA1 rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

Expression of plumECD.Scer\UAS.T:Mmus\Cd8a,T:Zzzz\FLAG under the control of Scer\GAL4Tab2-201Y rescues the γ neuron pruning defects seen in plumΔ1 mushroom body neuroblast clones.

Expression of plumScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Tab2-201Y in mushroom body neuroblast clones is sufficient to rescue the γ neuron pruning defects seen in plumΔ1 homozygous brains.
