Somatic gonadal precursors (SGPs) are specified normally as three clusters and primordial germ cells (PGCs) are associated with all three clusters in homozygous embryos. However, these clusters fail to adhere to each other at stage 13, and the mutant gonads remain elongated or split at stage 16 in the mutant embryos. Some mis-migrating PGCs are also seen outside the gonad from stage 13 onwards.

The number of SGP protrusions appear to be normal in mutant gonads at stage 15-16, extending between the PGCs as in wild type. The changes in SGP cell morphology and cell orientation that normally happen during gonad compaction do not occur in the mutant embryos.

Embryos that are derived from oskCE4/osk8 females and are zygotically homozygous for enaC14-06 show defects or delays in somatic gonadal precursor cluster adhesion at stage 13 and fail to complete gonad compaction, resulting in extended or split gonads at later stages, as occurs in enaC14-06 single mutant embryos.

67.8% of gonads do not form correctly in enaC14-06/+, shgg317/+ transheterozygous embryos, compared to 28.8% in enaC14-06/+ and 37.3% in shgg317/+ single heterozygotes.

More than 50% of gonads do not form correctly in enaC14-06/+, shg2/+ transheterozygous embryos.

51.9% of gonads fail to compact properly in enaC14-06/+, zip1/+ transheterozygous embryos, compared to 33.3% in zip1/+ single heterozygotes.

59.8% of gonads fail to compact properly in enaC14-06/+, arm1/+ transheterozygous embryos, compared to 22.1 in arm1/+ single heterozygotes.
