UASt regulatory sequences drive expression of Hsap\MYC isoform 1. The non-canonical CTG translation initiation codon of isoform 1 has been mutated to ATG. The alternative downstream ATG site has been mutated to ATC to prevent production of isoform 2 (which is usually produced using this translation start site) in this construct.
Females expressing Hsap\MYCScer\UAS.1 under the control of Scer\GAL4dm-PG45 show reduced viability, with greater survival at lower temperatures.
Clones in the wing disc expressing Hsap\MYCScer\UAS.1 under the control of Scer\GAL4Act5C.PP show a slight increase in cell size and a slight reduction in the fraction of cells in G[[1]] with a concomitant increase in cells in S or G[[2]] phase compared to controls. The clones show high levels of apoptosis.
